Description of the medicine: Thioguanin (Thioguaninum)

Thioguaninum. 2-Amino-6-mercaptopurine.

Synonyms: Lanvis, Lanvis, NSC-752, Thioguanine.

Yellowish or greenish-yellow crystalline powder. Almost nerastvorim in water and alcohol.

According to the chemical structure and mechanism of the antitumor effect, it is close to mercaptopurine, from which it differs only in the presence of an amino group in position 2.

Similarly, mercaptopurine is a purim antimetabolite.

The drug is slowly and not completely absorbed from the gastrointestinal tract. Peak concentrations in blood plasma observed after 6 - 8 hours after admission. About 40% is excreted in the urine within 24 hours (mainly as a 5-methylated metabolite).

Applied mainly with combined therapy of acute and chronic leukemia. It is especially effective in combination with cytarabine (see) and prednisolone, with acute and chronic myeloid leukemia (in the pre-blast stage and with blast crista). Can also be used in combination with antitumor antibiotics.

Take inside daily for 2 - 3 mg / kg, (daily dose of not more than 3 mg / kg). Duration of the course of treatment from 5 to 30 days, depending on the mode of application, efficacy, tolerability. When combined therapy (with cytarabine, antibiotics) daily dose is 2 - 2, 5 mg / kg; Course of treatment 5 days.

When using thioguanine, the same side effects are possible, as with the appointment of mercaptopurine. During the administration of thioguanine and after the end of treatment (within 2 weeks), it is necessary to monitor the parameters of peripheral blood and bone marrow hematopoiesis at least once every 2 weeks. At least once a week, the function of the liver is examined.

Contraindications are the same as for mercaptopurine.
