Medication Description: Lomustin (Lomustin)
1- (2-Chloroethyl) -3-cyclohexyl-1-nitrosourea.
Synonym: SiiNU, CeeNu.
Creamy white crystalline powder. Poor soluble in water and alcohol, soluble in acetone, dimethyl sulfoxide and dimethylformamide. Thermally unstable, melts with decomposition.
Contains in the molecule, along with a fragment of the nitrosourea, chloroethyl radical.
Alkylates phosphate, amine, hydroxyl, sulfhydryl and carboxyl groups, causes cross-links and DNA breaks.
Rapidly absorbed in the digestive tract, C max is 1-4 h; characterized by the ability to easily penetrate the blood-brain barrier.
Applied with tumors of the brain, lung, gastrointestinal tract, kidney, Hodgkin's disease, malignant lymphoma, melanoma, myeloma, and lymphosarcoma.
Assign inwards in a dose of 100-130 mg per 1 m 2 of the body surface 1 time every 6 weeks or 75 mg / m 2 every 3 weeks.
Leukopenia, thrombocytopenia, nausea, vomiting, anorexia, stomatitis, alopecia, abnormal liver functions, menstrual disorders may occur.
Contraindications: hematopoietic disorders (myelosuppression), marked disorders of the liver and kidneys, pregnancy, breastfeeding.
Method of production: tablets of 0.04 g (40 mg) (N. 50); capsules of 0.01; 0.04 and 0.1 g (N. 20).
