Description of the medicine: Lomustin (Lomustin)
1- (2-Chloroethyl) -3-cyclohexyl-1-nitrosourea.
Synonym: SiNU, CeeNu.
A creamy white crystalline powder. Slightly soluble in water and alcohol, soluble in acetone, dimethyl sulfoxide and dimethylformamide. Thermally unstable, melts with decomposition.
Contains in the molecule, along with a fragment of nitrosourea, a chloroethyl radical.
Alkylates phosphate, amine, hydroxyl, sulfhydryl and carboxyl groups, causes crosslinking and DNA breaks.
Quickly absorbed in the digestive tract, C max is 1-4 hours; different ability to easily penetrate the blood-brain barrier.
Used for tumors of the brain, lungs, gastrointestinal tract, kidneys, lymphogranulomatosis, malignant lymphoma, melanoma, myeloma and lymphosarcoma.
Assign orally in a dose of 100-130 mg per 1 m 2 of the body surface once every 6 weeks or 75 mg / m 2 every 3 weeks.
Leukopenia, thrombocytopenia, nausea, vomiting, anorexia, stomatitis, alopecia, liver dysfunction, menstrual irregularities can be observed.
Contraindications: hematopoiesis (myelosuppression), severe hepatic and renal impairment, pregnancy, breast-feeding.
Method of release: tablets of 0.04 g (40 mg) (N. 50); capsules of 0.01; 0.04 and 0.1 g (N. 20).
