Synestrol is a synthetic compound - a derivative of stilbene.

By chemical structure differs from steroid estrogenic hormones, but on biological and medical properties is close to them. To the group of stilbene derivatives are also diethylstilbestrol, dimestrol and other synthetic estrogen preparations.

In the first years after production, these synthetic estrogens were widely used in medical practice. Later, in connection with obtaining more effective and better tolerated drugs (ethinyl estradiol, etc.), the use of synestrol and diethylstilbestrol became more limited.

Synestrol for estrogenic activity is equivalent to estrone: 1 mg synestrol corresponds to 10,000 units.

Indications for use in women are basically the same as indications for the use of estrone. Sometimes prescribed for hypertrophy and prostate cancer in men.

Enter intramuscularly, under the skin and inside; Intramuscularly and under the skin in oily solutions, inside - in tablets. When administered, sinestrol is rapidly absorbed; Not destroyed in the gastrointestinal tract.

With hypogenitalism, congenital amenorrhea and abruptly underdeveloped uterus, 0.001 - 0.002 g (1-2 mg) is administered intramuscularly or 0.002 g (2 mg) orally daily for 4 to 6 weeks or more. If there is an effect, then prescribe progesterone (5 mg per day intramuscularly) or pregnin (10 mg 3 times a day orally) for 6 to 8 days. If necessary, hormone therapy courses are repeated.

In secondary amenorrhea, 1 to 2 mg sinestrol per day is given for 15 to 20 days, then for 6 to 8 days, progesterone or pregnin is given at the indicated dose.

With hypo-oligomenorrhea, the drug is prescribed 1 mg intramuscularly or intravenously every day or every other day during the first half of the intermenstrual period; With infertility due to the underdevelopment of the uterus - 1 mg intramuscularly or 1 - 2 mg orally in the first 7 to 8 days after menstruation; When menopausal disorders are prescribed by the interior of 0.0005 - 0.001 g (0.5 - 1 mg). The duration of treatment is individualized (see Estron).

To reduce lactation in women in the postpartum period, appoint 0.001 g 2 times a day inwards or 1 ml 0.1% solution once a day intramuscularly for 2 days; For complete suppression of lactation - 0,002-0,003 g per day or 1 ml of 0.1% solution 2 times a day for 5 - 7 days.

In breast cancer in women over 60 years of age, 2% synestrol solution is used. Enter daily, starting with 1 ml per day, then the daily dose is gradually increased by 5 ml. By careful observation, an optimal dose is set, which is administered for a long time.
